I haven't found it to reduce nitrates when using at the appropriate dosage. I did find that dosing a 1/4 teaspoon of Seachem Safe (more concentrated equivalent of Prime, and cheaper too) to reduce 5ppm of nitrates in 4 cups, which is way over the dosage recommendations.

As for affecting the ammonia test, it has never given me a false reading when I test before a water change or a couple of days later.

An interesting note is the recommended dosage at temperatures above 84F. They recommend using half of the volume tank dosage.
