There seem to be two different treament schedules. I just split the difference and waited 2 days and 1 night before the second dose. Also, I ended up treating all my tanks, because I worried about cross-contamination. In retrospect, that may have been a bit extreme.

One paper calls for this treament schedule:

Treat with the lights off and increased aeration.
Perform a largish water change prior to treatment.
Treat once for 24 hours.
Do a largish water change and vacuum to remove any paralyzed worms in the substrate.
Return tank to normal lighting/feeding/cleaning cycle.
Treat again in 5-7 days after a water change. If you know the parasite you are treating and its life cycle adjust the timing for the second treatment accordingly.
Do another water change with gravel vacuum.
Return to normal schedule.
Treat a third time 1 - 2 weeks after your second treatment.
Do another good vacuuming with water change and consider your treatment complete.

NOTE: This guy calls for a dose at 13 ppm or 5 grams/100 gal which is way out-of-bounds

They both recommend that you turn the tank light off as light decreases the effectiveness of levamizole.

I hope your guppies pull through. Are they still eating? If so, you probably caught it in time.

Oh yes, they're still eating, lol.

And Lamm sent me that same paper that you put up in here. It seems straightforward enough. I just don't trust myself without supervision- I get really freaked out and worry I'm not doing it right! The only animal I can successfully worm is a cat, that someone else is holding.

Well, treatment went well, and we got a little surprise at the end, when we decided to clean out the gravel. Still going to continue the treatments, but now we've got someone else to treat.
